STUDIES ON APPRORIATE CONDIONTS FOR EXPRESSING PlUGT1 PROTEIN IN RECOMBINANT PICHIA PASTORIS SYSTEM

Based on the single factor experiment, the effects of pH values and temperatures on growth of GS115/pPIZɑA-PlUGT1, and the effects of methanol concentrations, pH values and temperature on PlUGT1 protein production were studied. The optimal growth conditions of GS115/pPIZɑA-PlUGT1 were as follows: pH 6.5 and 30℃. The optimal expression conditions of PlUGT1 protein were found to be 0.5% of methanol concentration, 30℃ and pH 5.0. In this expression conditions for 72 h culture, the protein concentration in cultural solution is 0.721 mg/mL.
