NPR1和Defensin双价抗病基因过量表达载体的构建及其对沙田柚的遗传转化

Construction of Bivalent Over-Expression Vector for the NPR1 and Defensin Genes and its Transformation in Shatianyou Pummelo (Citrus grandis (L.) Osbeck)

  • 摘要: 柑橘黄龙病是一种严重危害柑橘生产上的毁灭性病害,目前对该病尚无有效的防治方法.为了培育柑橘黄龙病抗性育种材料,本文分别从克里曼丁橘和荔枝品种三月红叶片中提取总DNA,根据已公布于NCBI中的基因序列设计引物,克隆获得了NPR1和Defensin基因序列。以pbi-121作为载体骨架,构建了NPR1和Defensin双价抗病基因过量表达载体(命名为ND-pbi121).通过根癌农杆菌介导的遗传转化,获得了6株PCR检测为阳性的导入了NPR1和Defensin基因的沙田柚植株.这一结果将为探究NPR1和Defensin基因的过量表达对柑橘黄龙病的抗性影响提供试材和技术基础.

     

    Abstract: Citrus Huanglongbing(HLB) is a major citrus disease worldwide, and there is no effective control method for this disease. To strengthen the use and selection of resistant breeding materials, the open reading frames of NPR1 and Defensin genes were cloned frome genome which extracted from Citrus clementina leaves and varieties of litchi red March leaf, and ligated into the binary vector pbi-121 . The primers were designed according to the published se- quences in NCBI. This experiment successfLly constructed bivalent over -expression vector for the NPR1 and Defensin Genes, named ND-pbi121. Six transgenic plants were obtained through genetic transfor- mation mediated by Agrobacterium tumefaciens. The results will provide the basis for the study of the effect of NPR1 and Defensin on the resistance of Citrus Huanglongbing.
