花生AhAO2基因融合表达、纯化与抗体制备

FUSION EXPRESSION, PURIFICATION AND POLYCLONAL ANTIBODY PREPARATION OF PEANUT AHAO2 GENE

  • 摘要: 研究构建AhAO2基因片段原核表达重组质粒pPROEX-Hta-AhAO2的实验结果显示,重组pPROEX-Hta-AhAO2表达质粒在E.coli BL21中高表达His-AhAO2融合蛋白,采用亲和层析与电泳纯化His-AhAO2免疫新西兰大白兔,获得抗His-AhAO2抗体,Western blot检测显示其只与His-AhAO2融合蛋白结合.实验成功制备了抗AhAO2特异抗体.

     

    Abstract: The prokaryotic expression plasmid pPROEX-Hta-AhAO2 was constructed. The results showed that the recombinant pPROEX-Hta-AhAO2 plasmid expressed high amount of His-AhAO2 fusion protein in E. coli BL21. An anti-AhAO2 polyclonal antibody was obtained by immunizing the New Zealand rabbit with His-AhAO2 fusion protein purified by affinity chromatography and electrophoresis. The antibody of AhAO2 binds specifically to His-AhAO2 fusion protein fragment in western blot analysis.
