The Effect of Targeted Polypeptide Functionalized Cationic Polymer Loaded with ZNF580 Plasmid on Proliferation of Endothelial Cells

Abstract
A star-shaped cationic polymer POSS-(PDMAEMA)8 (PPD) based on oligomeric sesquioxane (POSS) was prepared with atom transfer radical polymerization(ATRP). In order to prepare POSS-(PDMAEMA)8-PPEGMA-CREDVW (PPD-CREDVW) cationic polymer, the CREDVW peptide, which targets endothelial cells (EA.hy926), was grafted onto the end of PPD cationic polymer through the thiol-double bond click chemical reaction. When n(N)/n(P)=5, PPD-CREDVW cationic polymer loaded with pEGFP-ZNF580 (pDNA) plasmid could form PPD-CREDVW/pDNA gene composite micelle with a particle size of 125.6±3.31 nm and a zeta potential of 10.64±1.65 mV through self-assembly. Compared with PPD/pDNA gene complexes micelles, PPD-CREDVW/pDNA gene complexes micelles could be uptaken by cells, had strong capacity for gene-carrying and had low cytotoxicity. Thus it can significantly promote the transfection and proliferation of endothelial cells.
