Construction of Bivalent Over-Expression Vector for the NPR1 and Defensin Genes and its Transformation in Shatianyou Pummelo (Citrus grandis (L.) Osbeck)

  • Citrus Huanglongbing(HLB) is a major citrus disease worldwide, and there is no effective control method for this disease. To strengthen the use and selection of resistant breeding materials, the open reading frames of NPR1 and Defensin genes were cloned frome genome which extracted from Citrus clementina leaves and varieties of litchi red March leaf, and ligated into the binary vector pbi-121 . The primers were designed according to the published se- quences in NCBI. This experiment successfLly constructed bivalent over -expression vector for the NPR1 and Defensin Genes, named ND-pbi121. Six transgenic plants were obtained through genetic transfor- mation mediated by Agrobacterium tumefaciens. The results will provide the basis for the study of the effect of NPR1 and Defensin on the resistance of Citrus Huanglongbing.
