High Frequency Plant Regeneration System by Thin Cell Layer Culture in Curcuma soloensis Valeton

  • Using the stem base of regenerated plantlets of Curcuma soloensis as explants, the stem base was cut into 05~08 mm thin layers. The effects of different hormone combinations, the position of stem base and culture conditions on adventitious shoots formation were studied. The results showed that the maximum frequency of adventitious shoots formation achieved from the thin layers at the position of the first root, and 15 adventitious shoots from the thin layers of a regenerated plantlet were obtained on the medium of MS+3 mg/L 6-BA+02 mg/L NAA after 45 d culture under the light culture condition. Adventitious shoot grew into a complete regenerated plantlet on the medium of 1/2 MS+1 g/L activated carbon. Plantlets were acclimatized and transplanted into pots. More than 95% of the plants survived and grew to mature plants.
