ABC1类蛋白激酶AtSIAK在植物抗盐胁迫响应中的功能研究

FUNCTIONAL ANALYSIS OF SIAK GENE IN RESPONSE TO SALT STRESS IN ARABIDOPSIS

  • 摘要: 在拟南芥中克隆了一个编码ABC1 类蛋白激酶的基因SIAK (Salt Induction ABC1-like Kinase), RT-PCR检测SIAK基因在拟南芥的根、茎、叶、花、角果等组织中均有表达,与利用SIAK基因的启动子驱动GUS报告基因的研究结果一致. SIAK基因受不同胁迫(ABA、SA、MV和NaCl)处理均有明显的上调表达,其中在NaCl处理下最明显.在不同浓度NaCl处理下,35S::SIAK 表达植株比WT 和siak1 突变体的种子萌发率和子叶变绿率,表明AtSIAK基因参与抗盐胁迫的响应.

     

    Abstract: We have cloned SIAK (Salt Induction ABC1-like Kinase) gene, encoded an ABC1 -like protein kinase in Aradidopsis thaliana, the expression patterns of SIAK in Aradidopsis were detected by RT-PCR and GUS staining. SIAK expressed in all tissues including root, leaf, silique and seedling, and it expressed most in leaves. The transcription level of the SIAK gene was analyzed by semi-quantitive RT-PCR in ABA, SA, MV and NaCl stress conditions, the transcription level was up-regulated, especially in the 0.3 M NaCl stress. Moreover, 35S- SIAK over expresssion plants had higher rate of cotyledon greening and long primer root length than siak-1 mutant and Col-0 after the treatment of NaCl. These results suggested that AtSIAK is involved in salt stress response.
