白藜芦醇上调GDF-15调控PI3K/Akt/Bcl-2信号通路对急性心肌梗死大鼠的保护作用

The Myocardial Preservation Effect of Resveratrol by Up-Regulating GDF-15 to Activate PI3K/Akt/Bcl-2 Pathway in Acute Myocardial Infarction Rats

  • 摘要: 为考察白藜芦醇对心肌缺血的保护机制,采用冠状动脉前降支结扎再灌注制备大鼠心肌缺血再灌注(I/R)模型,运用试剂盒考察血清酶学指标、NBT染色考察心肌梗死率、蛋白质印迹法(Western blotting) 考察GDF-15、PI3K、Akt、p-Akt、Bcl-2、Bax、caspase-9和caspase-3的蛋白相对表达量。结果表明:白藜芦醇对心肌缺血再灌注大鼠的心肌保护作用与通过对GDF-15的上调来激活PI3K/Akt信号通路,抑制心肌细胞线粒体凋亡作用机制有关。

     

    Abstract: To investigate the mechanism of resveratrol protecting myocardial ischemia, the rat myocardial ischemia-reperfusion (I/R) model was prepared with ligation and reperfusion of the anterior descending coronary artery. The serum enzyme indexes were analyzed with the kits, myocardial infarction rates were measured with NBT staining, and the western blotting method was used to investigate the relative protein expression levels of GDF-15, PI3K, Akt, p-Akt, Bcl-2, Bax, caspase-9 and caspase-3. The results showed that the myocardial protective effect of resveratrol on myocardial ischemia-reperfusion rats was related to the upregulation of GDF-15 to activate PI3K/Akt signaling pathway and inhibit cardiomyocyte mitochondrial apoptosis.
